Supplementary figure 1 -
A) Ngn3 expression induces glucagon, but not insulin expression. Double immunofluorescence staining of E5 chick embryo sections electroporated with Ngn3 expression plasmid. Sections were stained with alpha-Ngn3 (green) and alpha-insulin (INS) or alpha-glucagon (GLU) (red). Expression of Ngn3 induces glucagon, but not insulin expression.
B) Immunofluorescence staining of E5 chick embryo sections electroporated with MafA and MafB. Sections were stained for alpha-MafA (green) and alpha-insulin (red). Scale bar = 10mM.
Supplementary figure 2 -
MafA/B chimeric proteins do not influence glucagon expression. Double immunofluorescence analysis of chimeric MafA/B proteins. E5 chick embryo sections represent electroporations with chimeric MafA/B proteins alone (top) or in the presence of Ngn3 (bottom). Sections were stained with alpha-Maf (green) and alpha-glucagon (red). Glucagon expression was only induced in the presence of Ngn3. Scale bar = 10mM.
