THE EFFECT OF A β-ADRENERGIC BLOCKER ON INSULIN RELEASE FROM THE ISOLATED PERFUSED PANCREAS OF OBESE MICE

It appears now well accepted that adrenergic receptors are mediators of pancreatic insulin release. Agents blocking the β-adrenergic receptors are capable of inhibiting partially and temporarily glucose-induced insulin secretion (Loubatieres, Mariani, Sorel & Savi, 1971). The precise mechanisms and actions, however, are still unknown. So far, the influence of a β-receptor blocker has mainly been attributed to the effect on catecholamine action (Gagliardino, Bellone, Doria, Sanchez & Pereyra, 1970). The present report deals with a direct effect of a β-adrenergic blocking agent on glucose and non-glucose mediated exaggerated increase in insulin of obese mice in vitro, without added adrenergic substances.

Lean litter controls and obese mice (ob-ob, Jackson Lab., U.S.A.) weighing 50–75 g were used. The pancreas was isolated in an analogous way to that already described for rats (Sussman, Vaughan & Timmer, 1966). Arterial flow was achieved by cannulation of the thoracic aorta.
