PROGESTERONE IN THE BODY FAT OF THE DAIRY COW

Progesterone (pregn-4-ene-3:20-dione) has recently been reported to be present in relatively high concentration in the body fat of the dairy cow at mid-cycle (McCracken, 1963). This report gives details of the relative quantities of progesterone in the body fat of animals at different stages of the oestrous cycle.

METHODS: Samples of fat were obtained from cows slaughtered at a known stage of the oestrous cycle. Each sample was deep-frozen, homogenized in the frozen state, treated with 5N-NaOH (10 ml./100 g. fat) and extracted twice with an equal volume of peroxide-free ether. The ether extract was washed with 1/10 vol. de-ionized water and evaporated to dryness at 60° under reduced pressure. The fatty residue was dissolved in 250 ml. 70% aqueous methanol at 35°, cooled to – 15°, and filtered in the cold (Allen, 1932). The filtrate was evaporated to dryness on a rotary film evaporator at 50° and the residue
